이 library는 Brunello Human CRISPR Knockout Pooled Library(Addgene #73178)의 gRNA들을 사용하면서, guide cassette 뒤쪽에 inverted T7 promoter를 추가하여 전장 유전체 수준의 optical perturbation screening이 가능하도록 설계되었습니다. 이 방법은 살아있는 세포에서 형광 현미경을 이용해 광학적 표현형을 관찰하고, 같은 세포 안에서 Nuclear In-Situ Sequencing (NIS-Seq) 기술을 통해 각각의 gRNA를 직접 확인할 수 있습니다. 즉, 현미경으로 관찰 가능한 어떤 표현형이든, 그리고 핵이 있는 어떤 세포 유형이든, CRISPR 기반의 유전자 교란 스크리닝에 적용할 수 있다는 의미입니다.
